Function
CD9 antigen is a glycoprotein expressed on the surface of developing B lymphocytes, platelets, monocytes, eosinophils, basophil, stimulated T lymphocytes and by neurons and glial cells in the peripheral nervous system. Protein exists in three forms, and is known to carry covalently linked fatty acids. It is involved in platelet activation, aggregation, in cell adhesion, cell motility and tumor metastasis. It regulates paranodal junction formation, and is also required for gamete fusion.

Detected in platelets (at protein level). Expressed by a variety of hematopoietic and epithelial cells.
Post-translational Modification
Palmitoylated at a low, basal level in unstimulated platelets. The level of palmitoylation increases when platelets are activated by thrombin (in vitro). The protein exists in three forms with molecular masses between 22 and 27 kDa, and is known to carry covalently linked fatty acids. Palmitoylation by ZDHHC2 regulates CD9 expression, association with other tetraspanin family proteins and function in cell adhesion.

Western blot analysis of CD9 on SW480 cell lysates with Rabbit anti-CD9 antibody (ER80402) at 1/1,000 dilution.
Lysates/proteins at 10 µg/Lane.
Predicted band size: 25 kDa
Observed band size: 20 kDa
Exposure time: 2 minutes;
12% SDS-PAGE gel.
Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (ER80402) at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/200,000 dilution was used for 1 hour at room temperature. -
