Function
Fibroblast growth factor-1 (FGF-1), also designated acidic FGF, and fibroblast growth factor-2 (FGF-2), also designated basic FGF, are members of a family of growth factors that stimulate proliferation of cells of mesenchymal, epithe-lial and neuroectodermal origin. Additional members of the FGF family include the oncogenes FGF-3 (Int2) and FGF-4 (hst/Kaposi), FGF-5, FGF-6, FGF-7 (KGF), FGF-8 (AIGF), FGF-9 (GAF) and FGF-10-FGF-23. Members of the FGF family share 30-55% amino acid sequence identity and similar gene structure and are capable of transforming cultured cells when overexpressed in transfected cells. Cellular receptors for FGFs are members of a second multigene family including four tyrosine kinases, designated Flg (FGFR-1), Bek (FGFR-L), TKF and FGFR-3.

Western blot analysis of FGF21 on human liver tissue lysates with Rabbit anti-FGF21 antibody (ET1704-04) at 1/2,000 dilution.
Lysates/proteins at 20 µg/Lane.
Predicted band size: 22 kDa
Observed band size: 22 kDa
Exposure time: 10 seconds; ECL: K1801;
4-20% SDS-PAGE gel.
Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (ET1704-04) at 1/2,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature.

Immunohistochemical analysis of paraffin-embedded mouse liver tissue with Rabbit anti-FGF21 antibody (ET1704-04) at 1/1,000 dilution.
The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (ET1704-04) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
