Function
Estrogen related receptor alpha (ERRα) is a nuclear receptor in the superfamily of ligand-regulated transcription factors and is a member of the NR3B orphan nuclear receptor subgroup (consisting of α, β and γ). ERRα plays a role in modulating the estrogen signaling pathway. In addition, the expression of ERRα has been shown to increase during fasting and cold exposure. ERRα may be important for regulating mitochondrial biogenesis and oxidative metabolism by acting directly on genes necessary for mitochondrial function. Mice lacking ERRα are unable to maintain their body temperature in the cold. ERRα may also be involved in the maintenance and formation of cartilage. This information could be useful in finding therapeutic agents for a variety of diseases affecting the joints.

Sequence Similarity
Belongs to the nuclear hormone receptor family. NR3 subfamily.
Post-translational Modification
Phosphorylation on Ser-19 enhances sumoylation on Lys-14 increasing repression of transcriptional activity.; Sumoylated with SUMO2. Main site is Lys-14 which is enhanced by phosphorylation on Ser-19, cofactor activation, and by interaction with PIAS4. Sumoylation enhances repression of transcriptional activity, but has no effect on subcellular location nor on DNA binding.; Reversibly acetylated. Acetylation by PCAF/KAT2 at Lys-129, Lys-138, Lys-160 and Lys-162 and PCAF/KAT2 decreases transcriptional activity probably by inhibiting DNA-binding activity; deacetylation involves SIRT1 and HDAC8 and increases DNA-binding.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (ET1610-14) at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature.

Flow cytometric analysis of Estrogen Related Receptor alpha was done on Hela cells. The cells were fixed, permeabilized and stained with the primary antibody (ET1610-14, 1/50) (red). After incubation of the primary antibody at room temperature for an hour, the cells were stained with a Alexa Fluor 488-conjugated Goat anti-Rabbit IgG Secondary antibody at 1/1000 dilution for 30 minutes.Unlabelled sample was used as a control (cells without incubation with primary antibody; black).

Immunohistochemical analysis of paraffin-embedded human brain tissue with Rabbit anti-Estrogen Related Receptor alpha antibody (ET1610-14) at 1/1,000 dilution.
The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) (high pressure) for 2 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (ET1610-14) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
