FUNCTION

The DCC (Deleted in Colorectal Carcinoma) gene encodes a single-pass transmembrane receptor that functions as a receptor for netrin, a key axon guidance molecule in the developing nervous system. DCC plays a critical role in mediating axon attraction by binding to netrin ligands, guiding neuronal growth cones during neural development. Additionally, DCC can associate with UNC5 family proteins to trigger axon repulsion, highlighting its dual role in axon guidance. Beyond its role in the nervous system, DCC acts as a dependence receptor. In the absence of netrin ligand, DCC induces apoptosis, suggesting its function as a tumor suppressor. DCC has long been implicated in colorectal cancer, and its loss or epigenetic silencing is frequently observed in various cancers, supporting its role in tumor suppression. Recent research has revived interest in DCC as a potential therapeutic target due to its ligand-dependent tumor suppressor activity.
