Function
Interleukin 13 (IL-13) is a protein that in humans is encoded by the IL13 gene. The secondary structural features of IL-13 are similar to that of Interleukin 4 (IL-4); however it only has 25% sequence identity to IL-4 and is capable of IL-4 independent signaling. IL-13 is a cytokine secreted by T helper type 2 (Th2) cells, CD4 cells, natural killer T cell, mast cells, basophils, eosinophils and nuocytes. Interleukin-13 is a central regulator in IgE synthesis, goblet cell hyperplasia, mucus hypersecretion, airway hyperresponsiveness, fibrosis and chitinase up-regulation. It is a mediator of allergic inflammation and different diseases including asthma, and atopic dermatitis.

Sandwich ELISA analysis of human IL-13 matched pair antibodies

Elisa assay was performed by coating wells of a 96-well plate with 50 µl per well of capture antibody (HA725013) diluted in carbonate/bicarbonate buffer, at a concentration of 2 µg/ml overnight at 4℃. Wells of the plate were washed, blocked with 150 µl 0.05% tween-20 1% BSA blocking buffer, and incubated with serial diluted Recombinant Human IL-13 protein (HA210908) starting from 4,000 pg/ml to 0 pg/ml and detect antibody (HA725014, Biotin, 0.2 µg/ml) for 1 hour at 30℃ with shaking. Then the plate was washed and incubated with 50 µl per well of SA-HRP for 0.5 hour at 30℃ with shaking. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark. The reaction was stopped with sulfuric acid and absorbances were read on a spectrophotometer at 450 nm.

Interpolated concentrations of native IL-13 in human PBMC cell culture supernatant.

PBMC cells were stimulated with 10 µg/ml PHA-M and incubated for 6 days. The concentrations of IL-13 were interpolated from the IL-13 standard curves and corrected for sample dilution. Undiluted samples are as follows: unstimulated 50% and stimulated 50%. The mean IL-13 concentration was determined to be 5,701 pg/ml in PHA-M stimulated PBMC cell culture supernatant and undetectable in the unstimulated PBMC control.
