Function
ILF3 (interleukin enhancer binding factor 3) or TCP80 (translational control protein 80), is a ubiquitously expressed nuclear protein that exists in a heterodimer with NF45. NF90 contains two DRBM (double-stranded RNA(dsRNA-)-binding) domains and one DZF domain and, in association with NF45 primarily participates in the regulation of IL-2 expression by binding to the antigen receptor response element (ARRE) target sequence of the IL-2 enhancer. In neuronal cells, the NF45/NF90 heterodimer can repress human rhinovirus type 2 replication by binding to a 5' untranslated region of the viral RNA that encodes the internal ribosome entry site (IRES). NF45 and NF90 belong to the double-stranded RNA-binding protein family and both are substrates for the dsRNA-activated protein kinase, PKR. Due to alternative splicing events, six isoforms exist for NF90, namely NFAR-2 (or ILF3-E), NFAR-1 (or DRBP76), isoform 3, DRBP76α (or ILF3-A), DRBP76δ (also known as DRBP76γ or ILF3-C) and isoform 6.

This phosphorylation results in the dissociation of ILF2 from the ILF2-ILF3 complex resulting in a cytoplasmic sequestration of ILF3 where it can bind to viral RNAs and impede viral replication.; Methylated by protein arginine N-methyltransferase 1.

Western blot analysis of ILF3 on different lysates with Rabbit anti-ILF3 antibody (ET7108-49) at 1/500 dilution.
Lane 1: Rat heart tissue lysate(20 µg/Lane)

10% SDS-PAGE gel.
Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (ET7108-49) at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 200,000 dilution was used for 1 hour at room temperature. -
Immunocytochemistry analysis of A431 cells labeling ILF3 with Rabbit anti-ILF3 antibody (ET7108-49) at 1/50 dilution.
Cells were fixed in 4% paraformaldehyde for 10 minutes at 37 ℃, permeabilized with 0.05% Triton X-100 in PBS for 20 minutes, and then blocked with 2% negative goat serum for 30 minutes at room temperature. Cells were then incubated with Rabbit anti-ILF3 antibody (ET7108-49) at 1/50 dilution in 2% negative goat serum overnight at 4 ℃. Alexa Fluor®488 conjugate-Goat anti-Rabbit IgG was used as the secondary antibody at 1/1,000 dilution.Nuclear DNA was labelled in blue with DAPI. -
Immunocytochemistry analysis of SH-ST5Y cells labeling ILF3 with Rabbit anti-ILF3 antibody (ET7108-49) at 1/50 dilution.

Flow cytometric analysis of ILF3 was done on K562 cells. The cells were fixed, permeabilized and stained with the primary antibody (ET7108-49, 1/50) (purple). After incubation of the primary antibody at room temperature for an hour, the cells were stained with a Alexa Fluor 488-conjugated Goat anti-Rabbit IgG Secondary antibody at 1/1000 dilution for 30 minutes.Unlabelled sample was used as a control (cells without incubation with primary antibody; yellow).
