Function
CBX7 (Chromobox protein homolog 7) is a 251 amino acid nuclear protein that contains one N-terminal chromo domain and one C-terminal Pc box. Highly expressed in kidney, brain, heart and skeletal muscle, with weaker expression in peripheral blood leukocytes, CBX7 functions as a component of the chromatin-associated polycomb complex (PcG) and is involved in maintaining the transcriptionally repressed state of target genes. Additionally, CBX7 modifies chromatin and is thought to extend the cellular life span of epithelial cells by repressing p14 ARF expression, while simultaneously repressing telomerase activity. Due to its ability to repress the transcription of cell-cycle related proteins, CBX7 is thought to play a role in tumorigenesis, specifically in the development of follicular lymphoma and thyroid cancer.

Western blot analysis of cbx7 on A431 cell lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (ET1705-67, 1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1:200,000 dilution was used for 1 hour at room temperature.

Flow cytometric analysis of cbx7 was done on Hela cells. The cells were fixed, permeabilized and stained with the primary antibody (ET1705-67, 1/50) (red). After incubation of the primary antibody at room temperature for an hour, the cells were stained with a Alexa Fluor 488-conjugated Goat anti-Rabbit IgG Secondary antibody at 1/1000 dilution for 30 minutes.Unlabelled sample was used as a control (cells without incubation with primary antibody; black).
