Function
CD45R (also known as B220) is an isoform of CD45 (PTPRC), a member of the protein tyrosine phosphatase (PTP) family with an apparent molecular weight of 180–240 kDa due to extensive glycosylation. CD45R is expressed throughout B cell development, from pro-B to mature B cells, and is therefore widely used as a pan-B cell marker in mice. However, it is not entirely B cell-specific, as it is also expressed on subsets of activated T cells, NK cells, and plasmacytoid dendritic cells. CD45R is also found on abnormal T cell populations in autoimmune-prone mouse models such as MRL-Faslpr and MRL-Fasgld mice. Functionally, CD45 plays a critical role in antigen receptor signaling by regulating Src family kinases (e.g., Lck and Fyn) through its tyrosine phosphatase activity, rather than acting via classical ligand–receptor interactions.

This data was developed using HA601385, the same antibody clone in a different buffer formulation.
Immunohistochemical analysis of paraffin-embedded mouse spleen tissue with Rat anti-Mouse CD45R/B220 antibody (HA601385) at 1/1,000 dilution.
The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HA601385) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Flow cytometric analysis of mouse spleen cells labeling Mouse CD45R/B220 (HA601385).
Cells were washed twice with cold PBS and resuspend. Then stained with the primary antibody (HA601385, 1/1,000). After incubation of the primary antibody at +4℃ for an hour, the cells were stained with a Dylight 488 conjugate-Goat anti-Rat IgG Secondary antibody at 1/1,000 dilution for 30 minutes at +4℃.
