FUNCTION

CD8 is a pivotal integral membrane glycoprotein that is crucial for immune responses. In T-cells, CD8 primarily acts as a coreceptor for the MHC class I molecule:peptide complex. It simultaneously interacts with the T-cell receptor (TCR) and MHC class I proteins presented by antigen-presenting cells (APCs). This interaction recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling pathways. These pathways lead to lymphokine production, motility, adhesion, and activation of cytotoxic T-lymphocytes (CTLs), enabling CTLs to recognize and eliminate infected and tumor cells. In NK-cells, CD8A homodimers on the cell surface provide a survival mechanism. They facilitate conjugation and lysis of multiple target cells and promote the survival and differentiation of activated lymphocytes into memory CD8 T-cells. Overall, CD8 is indispensable in both T-cell mediated cellular immunity and NK-cell mediated natural immunity, offering strong protection against pathogens and internal anomalies.
