Function
Human influenza hemagglutinin (HA) is a surface glycoprotein required for the infectivity of the human virus. The HA tag is derived from the HA molecule corresponding to amino acids 98-106 has been extensively used as a general epitope tag in expression vectors. Many recombinant proteins have been engineered to express the HA tag, which does not appear to interfere with the bioactivity or the biodistribution of the recombinant protein. This tag facilitates the detection, isolation, and purification of the proteins.

Immunocytochemistry analysis of 293T cells labeling HA tag with Rabbit anti-HA tag antibody (HA720224F) at 1/100 dilution.
293T cells, transfected with empty control (top, negative) / HA-tagged CLK4 (middle, positive) / HA-tagged Nanos homolog 3 (bottom, positive) expression vector, respectively, were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 15 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature. Cells were then incubated with Rabbit anti-HA tag antibody (HA720224F, iFluor™ 594, red) at 1/100 dilution in 1% BSA in PBST overnight at 4 ℃. Nuclear DNA was labelled in blue with DAPI.
Flow cytometric analysis of 293T cells (left) / 293T cells transfected with HA-tagged CLK4 (right) labeling HA tag (HA720224F, iFluor™ 594).
Cells were fixed and permeabilized. Then incubated for 1 hour at +4℃ with HA tag (HA720224F, iFluor™ 594, red, 1/1,000) and Rabbit IgG Isotype Control (iFluor™ 594, green, 1/1,000). Unlabelled sample was used as a control (cells without incubation with primary antibody; black).
