Function
Ca2+-dependent actin filament-severing protein that has a regulatory function in exocytosis by affecting the organization of the microfilament network underneath the plasma membrane. Severing activity is inhibited by phosphatidylinositol 4,5-bis-phosphate (PIP2) (By similarity). In vitro, also has barbed end capping and nucleating activities in the presence of Ca2+. Required for megakaryocyte differentiation, maturation, polyploidization and apoptosis with the release of platelet-like particles. Plays a role in osteoclastogenesis (OCG) and actin cytoskeletal organization in osteoclasts. Regulates chondrocyte proliferation and differentiation. Inhibits cell proliferation and tumorigenesis. Signaling is mediated by MAPK, p38 and JNK pathways.

Western blot analysis of SCIN on different lysates with Rabbit anti-SCIN antibody (HA722465) at 1/1,000 dilution.
Lane 1: HEK-293 cell lysate (20 µg/Lane)

4-20% SDS-PAGE gel.
Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (HA722465) at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature.
