Function
Glucagon-like peptide-1 (GLP-1) is a 30- or 31-amino-acid-long peptide hormone deriving from tissue-specific posttranslational processing of the proglucagon peptide. It is produced and secreted by intestinal enteroendocrine L-cells and certain neurons within the nucleus of the solitary tract in the brainstem upon food consumption. The initial product GLP-1 (1–37) is susceptible to amidation and proteolytic cleavage, which gives rise to the two truncated and equipotent biologically active forms, GLP-1 (7–36) amide and GLP-1 (7–37). Active GLP-1 protein secondary structure includes two α-helices from amino acid position 13–20 and 24–35 separated by a linker region.

Wells of the plate were washed, blocked with 150 µl 0.05% tween-20 1% BSA blocking buffer, and incubated with serial diluted Recombinant Human GLP-1 protein starting from 2,000 pg/ml to 0 pg/ml and detect antibody (<a href="/products/HA725354" style="font-weight: bold;text-decoration: underline;">HA725354</a>, Biotin, 0.5 µg/ml) for 1 hour at 30℃ with shaking. Then the plate was washed and incubated with 100 µl per well of SA-HRP for 0.5 hour at 30℃ with shaking. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark. Elisa assay was performed by coating wells of a 96-well plate with 100 µl per well of capture antibody (HA725353) diluted in carbonate/bicarbonate buffer, at a concentration of 5 μg/ml overnight at 4℃. Wells of the plate were washed, blocked with 150 µl 0.05% tween-20 1% BSA blocking buffer, and incubated with serial diluted Recombinant Human GLP-1 protein starting from 2,000 pg/ml to 0 pg/ml and detect antibody (HA725354, Biotin, 0.5 µg/ml) for 1 hour at 30℃ with shaking.
