Function
CIP2A, also known as Cancerous Inhibitor of PP2A, is a protein that acts as an inhibitor of protein phosphatase PP2A. By preventing the dephosphorylation of MYC, CIP2A promotes anchorage-independent cell growth and tumor formation, thereby stabilizing MYC in human malignancies. In collaboration with TOPBP1, CIP2A plays a crucial role in responding to genome instability caused by acentric chromosome fragments derived from shattered chromosomes within micronuclei, which are commonly found in cancer cells. When the micronuclear envelope breaks down—a process linked to chromothripsis—the CIP2A-TOPBP1 complex tethers chromosome fragments during mitosis. This action ensures that the fragments are segregated to a single daughter cell nucleus, facilitating their re-ligation with minimal scattering and loss.

Western blot analysis of CIP2A on different lysates with Rabbit anti-CIP2A antibody (HA751590) at 1/5,000 dilution.

4-20% SDS-PAGE gel.
Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (HA751590) at 1/5,000 dilution was used in primary antibody dilution (K1803) at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature. -
CIP2A was immunoprecipitated from 0.2 mg Jurkat cell lysate with HA751590 at 2 µg/10 µl beads. Western blot was performed from the immunoprecipitate using HA751590 at 1/5,000 dilution. HRP Conjugated Anti-Rabbit IgG for IP Nano-secondary antibody at 1/50,000 dilution was used for 1 hour at room temperature.
