The LIN-12/Notch family of transmembrane receptors is believed to play a central role in development by regulating cell fate decisions. Ligands for Notch include Jagged1, Jagged2 and Delta. Jagged is a membrane protein and can activate Notch and prevent myoblast differentiation by inhibiting the expression of muscle regulatory and structural genes. It is involved in mammalian cardiovascular development and in cell-fate decisions during hematopoiesis. Jagged is expressed in adult and fetal tissues, and expression is upregulated in cervical squamous cell carcinoma. Familial Tetralogy of Fallot, the most common form of complex congenital heart disease, is caused by a mutation in the Jagged1 gene.

Tissue Specificity
Widely expressed in adult and fetal tissues. In cervix epithelium expressed in undifferentiated subcolumnar reserve cells and squamous metaplasia. Expression is up-regulated in cervical squamous cell carcinoma. Expressed in bone marrow cell line HS-27a which supports the long-term maintenance of immature progenitor cells.
Immunocytochemistry analysis of Raw264.7 cells labeling Jagged 1 with Rabbit anti-Jagged 1 antibody (R1706-10) at 1/50 dilution.
Cells were fixed in 4% paraformaldehyde for 10 minutes at 37 ℃, permeabilized with 0.05% Triton X-100 in PBS for 20 minutes, and then blocked with 2% negative goat serum for 30 minutes at room temperature. Cells were then incubated with Rabbit anti-Jagged 1 antibody (R1706-10) at 1/50 dilution in 2% negative goat serum overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (iFluor™ 488, HA1121) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI.
