Target
Function
This gene is a member of the TNF-receptor superfamily. The encoded protein is a receptor on antigen-presenting cells of the immune system and is essential for mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. AT-hook transcription factor AKNA is reported to coordinately regulate the expression of this receptor and its ligand, which may be important for homotypic cell interactions. Adaptor protein TNFR2 interacts with this receptor and serves as a mediator of the signal transduction. The interaction of this receptor and its ligand is found to be necessary for amyloid-beta-induced microglial activation, and thus is thought to be an early event in Alzheimer disease pathogenesis. Mutations affecting this gene are the cause of autosomal recessive hyper-IgM immunodeficiency type 3 (HIGM3). Multiple alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported.

Western blot analysis of CD40 on different lysates with Mouse anti-CD40 antibody (EM1901-41) at 1/1,000 dilution.
Lane 1: Raji cell lysate
Lane 2: Jurkat cell lysate
Lane 3: Daudi cell lysate
Lysates/proteins at 20 µg/Lane.
Exposure time: 37 seconds; ECL: K1802
Blocking: 5% NFDM/TBST, 1 hour at room temperature
Primary antibody: EM1901-41, 1/1,000 in 5% NFDM/TBST, overnight at 4 ℃
Secondary antibody: Goat Anti-Mouse IgG-HRP(HA1006), 1/50,000 in 5% NFDM/TBST, 1 hour at room temperature
Predicted band size: 31 kDa

Immunohistochemical analysis of paraffin-embedded human spleen tissue with Mouse anti-CD40 antibody (EM1901-41) at 1/200 dilution.
The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (EM1901-41) at 1/200 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX. -
