Function

Cell surface transmembrane glycoprotein CD200 receptor 1 is a protein that in humans is encoded by the CD200R1 gene. CD200R1 is expressed on the surface of myeloid cells and CD4+ T cells. It interacts with CD200 transmembrane glycoprotein that can be expressed on variety of cells including neurons, epithelial cells, endothelial cells, fibroblasts, and lymphoid cells. CD200R1 activation regulates the expression of pro-inflammatory molecules such as tumor necrosis factor (TNF-alpha), interferons, and inducible nitric oxide synthase (iNOS). This gene encodes a receptor for the OX-2 membrane glycoprotein. Both the receptor and substrate are cell surface glycoproteins containing two immunoglobulin-like domains. This receptor is restricted to the surfaces of myeloid lineage cells and the receptor-substrate interaction may function as a myeloid downregulatory signal. Mouse studies of a related gene suggest that this interaction may control myeloid function in a tissue-specific manner. Alternative splicing of this gene results in multiple transcript variants.

  • Sandwich ELISA analysis of Human CD200R1 matched pair antibodies<br /><br />Elisa assay was performed by coating wells of a 96-well plate with 100 µl per well of capture antibody (<a href="/products/HA722256" style="font-weight: bold;text-decoration: underline;">HA722256</a>) diluted in carbonate/bicarbonate buffer, at a concentration of 2 µg/mL overnight at 4℃. Wells of the plate were washed, blocked with 150 µl 0.05% tween-20 1%BSA blocking buffer, and incubated with serial diluted Recombinant Human CD200R1 protein (<a href="/products/HA210572" style="font-weight: bold;text-decoration: underline;">HA210572</a>) starting from 3,500 pg/ml to 0 pg/ml and detect antibody (<a href="/products/HA722257" style="font-weight: bold;text-decoration: underline;">HA722257</a>, Biotin, 0.2 µg/ml) for 1 hour at 30℃ with shaking. Then the plate was washed and incubated with 100 µl per well of SA-HRP for 0.5 hour at 30℃ with shaking. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark. The reaction was stopped with sulfuric acid and absorbances were read on a spectrophotometer at 450 nm.

Then the plate was washed and incubated with 100 µl per well of SA-HRP for 0.5 hour at 30℃ with shaking. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark.   • The concentrations of CD200R1 were interpolated from the CD200R1 standard curves and corrected for sample dilution.Undiluted samples are mixed serum from twenty-four volunteers 25%. The mean CD200R1 concentration was determined to be 3,702 pg/mL in mixed serum from twenty-four volunteers.
