Forkhead box protein G1 is a protein that in humans is encoded by the FOXG1 gene. This gene belongs to the forkhead family of transcription factors that is characterized by a distinct forkhead domain. The complete function of this gene has not yet been determined; however, it has been shown to play a role in the development of the brain and telencephalon. Mutations of FOXG1 are the cause of FoxG1 syndrome. FoxG1 syndrome is characterized by microcephaly and brain malformations. It affects most aspects of development and can cause seizures. FOXG1 syndrome is classified as an autism spectrum disorder and was previously considered a variant of Rett syndrome.

Western blot analysis of FOXG1 on different lysates with Rabbit anti-FOXG1 antibody (HA723067) at 1/2,000 dilution.
Lane 1: Neuro-2a cell lysate (20 µg/Lane) Lane 2: Mouse brain tissue lysate (40 µg/Lane)
Predicted band size: 52 kDa Observed band size: 58 kDa Exposure time: Lane 1: 10 seconds; Lane 2: 3 minutes; ECL: K1801; 4-20% SDS-PAGE gel.
Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (HA723067) at 1/2,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature.
FOXG1 was immunoprecipitated from 0.2 mg Neuro-2a cell lysate with HA723067 at 2 µg/10 µl beads. Western blot was performed from the immunoprecipitate using HA723067 at 1/2,000 dilution. Anti-Rabbit IgG for IP, AlpSdAbs® VHH(HRP) (025-100-005) at 1/5,000 dilution was used for 1 hour at room temperature.
