Catalyzes the translocation of specific phospholipids from the cytoplasmic to the extracellular/lumenal leaflet of membrane coupled to the hydrolysis of ATP. Transports preferentially phosphatidylserine over phosphatidylcholine 2. Plays a role in lipid homeostasis and macrophage-mediated phagocytosis. Binds APOA1 and may function in apolipoprotein-mediated phospholipid efflux from cells 7, 8, 9. May also mediate cholesterol efflux. May regulate cellular ceramide homeostasis during keratinocyte differentiation.Involved in lipid raft organization and CD1D localization on thymocytes and antigen-presenting cells, which plays an important role in natural killer T-cell development and activation (By similarity). Plays a role in phagocytosis of apoptotic cells by macrophages (By similarity). Macrophage phagocytosis is stimulated by APOA1 or APOA2, probably by stabilization of ABCA7 (By similarity). Also involved in phagocytic clearance of amyloid-beta by microglia cells and macrophages (By similarity). Further limits amyloid-beta production by playing a role in the regulation of amyloid-beta A4 precursor protein (APP) endocytosis and/or processing. Amyloid-beta is the main component of amyloid plaques found in the brains of Alzheimer patients.

Western blot analysis of ABCA7 on different lysates with Rabbit anti-ABCA7 antibody (HA723974) at 1/5,000 dilution.
Lane 1: Neuro-2a cell lysate (no heat) (20 µg/Lane)
Lane 2: Mouse brain tissue lysate (no heat) (40 µg/Lane)
Lane 3: Mouse cerebellum (P7) tissue lysate (40 µg/Lane)
Notice: no heat means the lysate is not boiled.
Predicted band size: 234 kDa
Observed band size: 250 kDa
Exposure time: 3 minutes; ECL: K1801;
4-20% SDS-PAGE gel.
Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (HA723974) at 1/5,000 dilution was used in primary antibody dilution (K1803) at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature. -
Immunohistochemical analysis of paraffin-embedded mouse thymus tissue with Rabbit anti-ABCA7 antibody (HA723974) at 1/200 dilution.
The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HA723974) at 1/200 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX. -
