Collagen, type I, alpha 1, also known as alpha-1 type I collagen, is a protein that in humans is encoded by the COL1A1 gene. COL1A1 encodes the major component of type I collagen, the fibrillar collagen found in most connective tissues, including cartilage. Collagen is a protein that strengthens and supports many tissues in the body, including cartilage, bone, tendon, skin and the white part of the eye (sclera). The COL1A1 gene produces a component of type I collagen, called the pro-alpha1(I) chain. This chain combines with another pro-alpha1(I) chain and also with a pro-alpha2(I) chain (produced by the COL1A2 gene) to make a molecule of type I procollagen. These triple-stranded, rope-like procollagen molecules must be processed by enzymes outside the cell. Once these molecules are processed, they arrange themselves into long, thin fibrils that cross-link to one another in the spaces around cells. The cross-links result in the formation of very strong mature type I collagen fibers. Collagenous function includes rigidity and elasticity.

Sandwich ELISA analysis of Human Pro-COL1A1 matched pair antibodies

Detector: HA724510, Human/Monkey Pro-COL1A1 Rabbit mAb [PSH25-16]
Elisa assay was performed by coating wells of a 96-well plate with 100 µl per well of capture antibody (HA724509) diluted in carbonate/bicarbonate buffer, at a concentration of 2 μg/mL overnight at 4℃. Wells of the plate were washed, blocked with 150 µl 0.05% tween-20 1% BSA blocking buffer, and incubated with serial diluted Recombinant Human Pro-COL1A1 protein (HA211375) starting from 2,500 pg/mL to 0 pg/mL and detect antibody (HA724510, Biotin, 0.2 μg/mL) for 1 hour at 30℃ with shaking. Then the plate was washed and incubated with 100 µl per well of SA-HRP for 0.5 hour at 30℃ with shaking. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark. The reaction was stopped with sulfuric acid and absorbances were read on a spectrophotometer at 450 nm. -

Interpolated concentration of native Pro-COL1A1 was measured in duplicate at different sample concentrations and interpolated from the Pro-COL1A1 standard curves. The interpolated dilution factor corrected values were plotted (mean +/- SD, n=2). The mean Pro-COL1A1 concentration was determined to be 122,731 pg/mL in rhesus monkey serum and 25,357 pg/mL in human serum. -
