Target
Function
C3 plays a central role in the activation of the complement system. Its processing by C3 convertase is the central reaction in both classical and alternative complement pathways. After activation C3b can bind covalently, via its reactive thioester, to cell surface carbohydrates or immune aggregates. Derived from proteolytic degradation of complement C3, C3a anaphylatoxin is a mediator of local inflammatory process. It induces the contraction of smooth muscle, increases vascular permeability and causes histamine release from mast cells and basophilic leukocytes. Defects in C3 are the cause of complement component 3 deficiency (C3D). Genetic variation in C3 is associated with susceptibility to age-related macular degeneration type 9 (ARMD9). Defects in C3 are a cause of susceptibility to hemolytic uremic syndrome atypical type 5 (AHUS5).

Wells of the plate were washed, blocked with 150 µl 0.05% tween-20 1% BSA blocking buffer, and incubated with serial diluted Recombinant Mouse Complement C3 protein (<a href="/products/HA211093" style="font-weight: bold;text-decoration: underline;">HA211093</a>) protein starting from 1,000,000 pg/ml to 0 pg/ml and detect antibody (<a href="/products/HA725144" style="font-weight: bold;text-decoration: underline;">HA725144</a>, HRP, 0.5 µg/ml) for 1 hour at 30℃ with shaking. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark. Elisa assay was performed by coating wells of a 96-well plate with 50 µl per well of capture antibody (HA725143) diluted in carbonate/bicarbonate buffer, at a concentration of 0.5 µg/ml overnight at 4℃. Wells of the plate were washed, blocked with 150 µl 0.05% tween-20 1% BSA blocking buffer, and incubated with serial diluted Recombinant Mouse Complement C3 protein (HA211093) protein starting from 1,000,000 pg/ml to 0 pg/ml and detect antibody (HA725144, HRP, 0.5 µg/ml) for 1 hour at 30℃ with shaking. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark. The reaction was stopped with sulfuric acid and absorbances were read on a spectrophotometer at 450 nm.
