Function
Phospholipase C, gamma 1, also known as PLCG1 and PLCgamma1, is a protein that in humans involved in cell growth, migration, apoptosis, and proliferation. It is encoded by the PLCG1 gene and is part of the PLC superfamily. PLCγ1 is a cell growth factor from the PLC superfamily. PLCγ1 is used during the cell growth and in a cell migration and apoptosis, all of which are vital cell processes that, if disrupted by mutations, can cause cancerous cells to form within the body. Mutations in this protein show an increase in issues in cells regarding regulation of proliferation and their cell signaling. PLCγ1 roles are also involved in neuronal actin growth, calcium signaling, and brain development. It is highly regulated by multiple factors, such as PIK3, AMPK, and FAK. It is part of the PIP3 pathway and leads to and increase in calcium in the cells. In neuronal cells, PLCγ1 is highly involved in actin cytoskeleton organization and synaptic plasticity.

Western blot analysis of Phospho-PLC-gamma-1 (S1248) on different lysates with Rabbit anti-Phospho-PLC-gamma-1 (S1248) antibody (HA751481) at 1/2,000 dilution.
Lane 1: A431 cell lysate
Lane 2: A431 starved overnight then add 20ng/mL EGF for 5 minutes cell lysate
Lane 3: A431 starved overnight then add 20ng/mL EGF for 5 minutes cell lysate, then the membrane treated with λpp for 1 hour
Lysates/proteins at 20 µg/Lane.
Predicted band size: 149 kDa
Observed band size: 149 kDa
Exposure time: 59 seconds; ECL: K1801;
4-20% SDS-PAGE gel.
Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (HA751481) at 1/2,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature. -

Phospho-PLC-gamma-1 (S1248) was immunoprecipitated from 0.2 mg A431 starved overnight then add 20ng/mL EGF for 5 minutes cell lysate with HA751481 at 2 µg/10 µl beads. Western blot was performed from the immunoprecipitate using HA751481 at 1/2,000 dilution. HRP Conjugated Anti-Rabbit IgG for IP Nano-secondary antibody at 1/5,000 dilution was used for 1 hour at room temperature.
Lane 1: A431 starved overnight then add 20ng/mL EGF for 5 minutes cell lysate (input)
Lane 2: HA751481 IP in A431 starved overnight then add 20ng/mL EGF for 5 minutes cell lysate
Lane 3: Rabbit IgG instead of HA751481 in A431 starved overnight then add 20ng/mL EGF for 5 minutes cell lysate
Blocking/Dilution buffer: 5% NFDM/TBST
