白细胞介素-4（IL-4），也称为B细胞刺激因子-1，是一种单体，约13 kDa-18 kDa Th2细胞因子，在免疫反应期间显示多效性作用。它是一种糖基化的多肽，含有三个链内二硫键，采用捆绑的四个α-螺旋结构。人IL-4由24个氨基酸信号序列合成。交替剪接生成具有 16个氨基酸内部缺失的亚型。成熟的人的IL-4分别与牛、小鼠和大鼠的IL-4共享55%、39%和43%的氨基酸序列同一性。人、小鼠和大鼠IL-4在其活动中具有物种特异性。
IL-4通过两个受体复合物发挥其作用。在造血细胞上表达的 I 型受体是结合 IL-4 R α和普通 γ 链（IL-2、-7、-9、-15 和 -21 受体的共享亚基）的配体的异二聚体。非造血细胞上的II型受体由IL-4 R α和IL-13 R α 1组成。II型受体也转导IL-13介导的信号。IL-4主要由Th2偏向的CD4+ T细胞、肥大细胞、嗜碱性粒细胞和嗜酸性粒细胞表达。它促进人B细胞的细胞增殖、存活和免疫球蛋白类转换为IgG4和IgE，通过幼稚CD4+ T细胞获得Th2表型、肥大细胞、嗜酸性粒细胞和嗜碱性粒细胞的启动和趋化性，以及上皮细胞的增殖和活化。IL-4在过敏性炎症和哮喘的发展中起主导作用。
